RPMI 1640 Media, heparinized syringe, or dark green top (sodium heparin) tube. Other anticoagulants are not recommended and are harmful to the viability of the cells.
2-3 mL bone marrow
1 mL bone marrow
Specimen transport should be arranged so that the specimen is received by MLabs the same day it is collected. Collect bone marrow using aseptic technique. Place 1 - 3 mL bone marrow aspirate in a dark green top (sodium heparin) tube. Invert the tube several times to prevent clotting. Send at room temperature as soon as possible. Bone core biopsies must be transported in sterile transport media. Do not allow specimen to overheat or freeze. Clotted samples, samples containing <5 million cells, and samples that are greater than 24 hours old are considered suboptimal.
RPMI 1640 Media tube. Sterile syringe rinsed with sodium heparin or green top vacutainer with sodium heparin. All pediatric bone marrow specimens and bone core biopsies must be transported in RPMI 1640 Media provided by the Cytogenetics laboratory and available from Specimen Processing. Do not allow specimen to overheat or freeze. Clotted samples, samples containing <5 million cells, and samples that are greater than 24 hours old are considered suboptimal. Please complete a MiChart test request and submit it with the specimen. A Cytogenetics Laboratory Requisition is available online at: http://www.pathology.med.umich.edu/handbook/Tables/UM_REQ_CYTOGENETICS.pdf.
Monday - Friday, 8:00am - 4:00pm. Call the laboratory at 734-763-5805 for additional information.

Short term cell culture and chromosome analysis, cultures with and without mitogens. Fluorescence In Situ Hybridization (FISH) when indicated. Samples with an indication of mature B-cell lymphoma including, but not limited to, CLL/SLL, follicular lymphoma, mantle cell lymphoma and marginal zone lymphoma, will have an extra culture inoculated with CpG-oligonucleotide (CpG) and Interleukin-2 (IL2). Similarly, samples with an indication of mature T-cell lymphoma/leukemia (T-PLL, AITCL, ALCL) will have an extra culture inoculated with Phytohemagglutinin (PHA). A minimum of 10 metaphase cells are analyzed from mitogen stimulated cultures, in addition to the standard 20 metaphase cells analyzed from the unstimulated cultures.
Interpretive report provided.
Identification of specific chromosomal abnormalities associated with hematologic disease. Bone Marrow specimens are most commonly sent as part of the evaluation of acute leukemia (lymphoid or myeloid), MDS, or myeloproliferative disorders including chronic myelogenous leukemia, in the latter case to look for a Philadelphia chromosome.
Cells must be capable of dividing.
Appropriate FISH testing will be performed at an additional charge when indicated. FISH testing is available for a number of oncology probes and panels, including but not limited to, BCR/ABL, PML/RARA, CBFB/MYH11, KMT2A, IGH/MYC, MYC, and MDS FISH and ALL panels, (see Cytogenetics, FISH for Malignancy). For additional information on FISH testing and available probes, contact the Cytogenetics Laboratory. By ordering this test the clinician acknowledges that additional reflex testing will be performed and billed at a separate additional charge if indicated.
